Dilutores liposoma y Tris sobre semen fresco y descongelado en la inseminación artificial en bovinos

En inseminación bovina, la yema de huevo en dilutores de semen aumenta la contaminación microbiana, requiriendo alternativas. El objetivo del estudio fue evaluar los dilutores liposoma (Optixcell) y Tris-yema de huevo (Tris-YH) sobre los parámetros espermáticos del semen fresco y descongelado, y tasa de preñez en vacas inseminadas. Se colectó semen de un toro, inmediatamente se diluyó con Optixcell o Tris-YH, se evaluó los parámetros espermáticos del semen fresco, se congeló convencionalmente, luego el semen descongelado local e importado fueron evaluados. El diagnóstico de gestación se realizó a los 45 días. Los parámetros espermáticos del semen fresco diluido con Optixcell y Tris-YH no mostraron diferencias significativas en motilidad total (p=0.98), motilidad progresiva (p=0.81), vitalidad (p=0.64), integridad de membrana plasmática (p=0.64) ni anormalidades espermáticas (p=0.36). En semen descongelado, tampoco hubo diferencias (p>0.05). Sin embargo, el semen descongelado importado presentó valores superiores de motilidad total (p=0.004), motilidad progresiva (p=0.006), vitalidad (p=0.006) e integridad de membrana plasmática (p= 0.002) frente al semen local. La tasa de preñez fue similar (p=0.83) entre vacas inseminadas con semen importado versus local. Se concluye que los dilutores Optixcell y Tris-YH no afectaron la calidad espermática del semen fresco y descongelado local, mientras que el semen importado presentó parámetros espermáticos superiores. Sin embargo, esto no se tradujo en diferencias en la tasa de preñez en vacas inseminadas.
In bovine artificial insemination, the use of egg yolk in semen extenders increases microbial contamination, making alternatives necessary. The objective of this study was to evaluate the effects of the liposome-based extender (Optixcell) and the Tris-egg yolk extender (Tris-YH) on sperm parameters of fresh and frozen-thawed semen, as well as the pregnancy rate in inseminated cows. Semen was collected from one bull, immediately diluted with Optixcell or Tris-YH, and evaluated for sperm parameters in the fresh state. Samples were then conventionally frozen, and both locally frozen-thawed and imported frozen-thawed semen were subsequently evaluated. Pregnancy diagnosis was performed 45 days after insemination. Fresh semen diluted with Optixcell and Tris-YH showed no significant differences in total motility (p=0.98), progressive motility (p=0.81), viability (p=0.64), plasma membrane integrity (p=0.64), or sperm abnormalities (p=0.36). Similarly, no significant differences were observed in frozen-thawed semen (p>0.05). However, imported frozen-thawed semen exhibited significantly higher total motility (p=0.004), progressive motility (p=0.006), viability (p=0.006), and plasma membrane integrity (p=0.002) compared to locally frozen-thawed semen. The pregnancy rate was similar (p=0.83) between cows inseminated with imported and local semen. It is concluded that the Optixcell and Tris-YH extenders did not affect the sperm quality of fresh or locally frozen-thawed semen, whereas imported semen showed superior sperm parameters. Nevertheless, these differences did not result in improved pregnancy rates in inseminated cows.
